This sounds OS dependent. I just use my headset's mute button.

Most operating systems have a sound settings panel, you can mute the microphone input there. I run linux, and I assume you're running windows.

If you use google hangouts there is an onscreen mute button, if you use skype you can mute with icon or set it up for push to talk. Depends on what you use for voice chat in your games.

There's even a mute button in the tokbox through roll20. You just have to have your display set to more than just "Names only" to get access to it.
