Is There A Way I Can Edit A Picture In Game?

I would just run it through Paint real quick. Copy the adjacent square and paste it over the E, save and then drop into Roll20. Alternately, you might try putting an art object over it if you can match the color.

If you are using donjon map gen (it looks like you are) there is an option for 'player map' on it to download without letters/numbers, edit: Or, well there use to be, just looked to find the name of the link and its missing them...last week I used it, it had the option to download the pdf, the playermap, and the DM map

You could simply draw a filled-in white square over the letters, I suppose.
