Previous experience with D&D.

Is it possible to play a game on Roll20 without ever playing a D&D game in person? Does it help to have played a D&D game in person?
Yes and Yes. I will add that at least one person needs to have some experience with the game or things will not go well. Preferably the DM.

It is very possible to play a game on roll20 without playing it in person. While it does help having experience with the system, it is not to hard to learn the game. The main thing you need is to have the books (physical item or digital) so you can read and follow the instructions. Learning the system is pretty the same in person or on roll20. Read the books, setup a simple game, get some friends together, and have FUN.

You can get by with the basic (free) rules for 5e, but they have much less content than the real deal, with only 114 page player basic guide and 64 page dungeon master guide. But I second Pat S.'s And Patrick's opinions.
I'd also suggest to have a look at some of the streams on twitch or youtube. There is a pretty awesome group (imho) playing the Tombs of Annihilation campaign: https://www.twitch.tv/videos/174013141 You can pick up a lot of the general "how to play d&d" from just watching those shows.
Just grab a friend or two and jump right in there! It's great fun. :)
there really is no wrong way to play D&D we just call that "house rules" but watching some of the games on youtube or what ever can show you what it can be. just have fun if its not fun your are doing it wrong for you

You can read the free ruleset online here for D&D 5e. The free rules have everything you need to build your first character. If you like the game, you may want to look into buying the Player's Handbook but the free rules is a great way to test out the system. Good luck with your first game!

The player's handbook is all you need to read for now. Get the starter, read it, find a group to game with (the LFG forum or The Game Search), and have fun. Be patient about getting into a group/game as it could take time for there are thousands of players looking for a game. 5e game search with welcome new players and free : I did not check the mature filter due to not knowing your age nor views on mature subjects/language, nor did I include any language other than English.
