Dynamic Light Line Thickness

Is there really a difference between which line thickness used? Aside from easier to connect walls with thicker lines, thereby preventing gaps, what other advantage does using a Small vs Medium vs Large line provide?

I don't believe there is any difference between different sized lines.

In LDL, vision is blocked from the center of the line, regardless of the line thickness used. In UDL, vision is blocked from the edge of the line, so a thicker line blocks more of the map. While thicker lines can make it easier to prevent games in UDL, the same effect could be achieved by adding "tails" to lines rather than just using a straight line. This also makes it easier to grab the line as sometimes the VTT loses the ability to grab short, straight lines.
